Abstract: This report validates a technique of separating and labeling canine platelets using indium-111-labeled 8-hydroxyquinoline (111In-8-HQ). Platelet survival time in six normal dogs was 6.3 +/- 0.7 (SD) days. Labeling efficiency using this technique was 51.8 +/- 9.2% (SEM) and elution loss of 111In-8-HQ label from platelets was consistently less than 4%. 111In label offers no observable advantages over the more traditional 51Cr label for platelet survival studies. However, the high efficiency labeling and -[gamma]-emission characteristics of 111In-8-HQ make it more suitable for non-invasive clot localization in patients with early pulmonary emboli and peripheral venous thrombi.
