Methodology
  • 1) RA (10 nM) was applied to organotypic cultures of the mouse organ of Corti (E14.5) for 7 days in vitro (DIV) and compared to the solvent control (DMSO). Specimens were fixed with 4% paraformaldehyde (PFA) and stained with anti-MYO7A and anti-SOX2 antibodies, then counterstained with DAPI. Images were captured using a laser scanning microscope at representative locations along the cochlear duct. Data represent raw data.

  • 2) RA (500 nM), the RA receptor blocker AGN 193109 (100 nM), and DMSO control were applied for 72 hours to organotypic cultures of the mouse organ of Corti (E14.5). Specimens were fixed with 4% PFA and stained using Lfng and Atoh1 fluorescence in situ hybridization probe sets, anti-SOX2 antibody, and counterstained with DAPI. Images were taken using a confocal microscope at representative locations along the cochlear duct.
Description
  • The aim of this study was to determine the role of retinoic acid (RA) in the development of the organ of Corti. Two ex vivo organ culture experiments were conducted, in which the sensory epithelium of the cochlea, the organ of Corti, was dissected and transferred to a culture dish. This model allows for the application of drugs or compounds in the culture media, enabling us to assess their effects using light microscopy. Two paradigms were tested.
