Methodology
  • Single molecule Förster resonance energy transfer (smFRET) experiments were performed on a prism-based TIRF microscope (IX 71; Olympus) with a 1.2-numerical aperture 60X water-immersion objective (Olympus UplanApo) and an intensified CCD camera (I-Pentamax, Princeton Instruments). The DY547 molecules were excited with a 532 nm diode laser and emission from DY547 and Cy5 were recorded at a 100 ms exposure time in mj2 format. Melting experiments were performed on a Beckman DU® 640B spectrophotometer fitted with a high performance temperature controller unit, transport accessory and Tm six-cell holder.
Description
  • This work contains the experimental data and associated analysis that are described in the research publication entitled "Protein unties the pseudoknot: S1-mediated unfolding of RNA higher order structure". This work contains multiple zip files, each of which represents one of the principal experiment groups presented in the publication. Each experiment group contains movie and analysis files corresponding to various experimental conditions related to that experiment group. A zip file containing the processed single molecule data files used as input for Single Molecule Cluster Analysis (SiMCAn), and the corresponding analysis outputs are also provided.

  • Additionally, there are zip files containing: the IDL and Matlab scripts that were used to process the single molecule data, Matlab scripts used to analyze melting curve data, and Matlab scripts and used to perform SiMCAn analysis.
