Methodology
  • This record contains two types of data: (1) images showing ancestry of alleles in recombinant backcross flies inferred from Multiplexed Shotgun Genotyping (MSG), and (2) video recordings of courtship for Drosophila elegans, Drosophila gunungcola, and progeny from crosses between these two species. For (1), MSG data was generated following protocols described in Andolfatto et al. (2011) Genome Research, 21(4), 610-617 and Cande et al., (2012) PLoS One, 7(8), e43888. For (2), flies were videotaped for 20 min using a Canon VIXIA HF R500 camcorder. Videos were recorded between 09:00 and 16:00 at 23ºC.

Key Points:
- We generated over 900 MP4 videos that capture divergent courtship behavior in backcross recombinant flies.

Research Overview:
To study the genetic basis of behavioral divergence between Drosophila elegans and D. gunungcola, we generated >900 MP4 videos that illustrate species differences in courtship behavior in individual recombinant male flies courting virgin D. elegans females. Courtship behavior is present in most but not all videos. We captured the videos using four Canon VIXIA HF R500 camcorders mounted to Manfrotto (MKCOMPACTACN-BK) aluminum tripods. Courtship chambers were designed based off of the specifications described in Simon, J. C., & Dickinson, M. H. (2010). A new chamber for studying the behavior of
Drosophila. Plos One, 5(1), e8793. Using these videos, we captured instances of wing display behavior to generate quantitative measurements for quantitative trait locus (QTL) mapping.
