Methodology
  • Action currents, which are associated with action potential firing, were recorded from GFP-identified neurons in acutely-prepared brain slices from control and PNA female mice at three weeks of age and adults. These events were detected, confirmed, and later analyzed with custom software written in Igor Pro. Prepubertal development of gonadotropin-releasing hormone neuron activity is altered by sex, age and prenatal androgen exposure. Endocrinology 2017; 158:3943-3953
Description
  • This data is a subset of that originally produced as part of an effort to characterize GnRH neuron activity during prepubertal development in control and PNA mice and investigate the potential influences of sex and PNA treatment on this process (1). It was later used in (2) to further investigate the firing patterns of GnRH neurons in these categories of mice and determine how these patterns might differ based on age and treatment condition. The data files can be opened and examined using Wavemetric's Igor Pro software. Code used to further examine and visualize the data can be found at  https://gitlab.com/um-mip/mc-project-code. This research was supported by National Institute of Health/Eunice Kennedy Shriver National Institute of Child Health and Human Development R01 HD34860 and P50 HD28934. (1) Dulka EA, Moenter SM. Research Overview:
This data is a subset of that originally produced as part of an effort to characterize GnRH neuron activity during prepubertal development in control and PNA mice and investigate the potential influences of sex and PNA treatment on this process (1). It was later used in (2) to further investigate the firing patterns of GnRH neurons in these categories of mice and determine how these patterns might differ based on age and treatment condition.

Methodology:
Action currents, which are associated with action potential firing, were recorded from GFP-identified neurons in acutely-prepared brain slices from control and PNA female mice at three weeks of age and adults. These events were detected, confirmed, and later analyzed with custom software written in Igor Pro. For more specific details on the data acquisition process, please refer to (1).

Software specifications:
Wavemetric's Igor Pro is required to open the data files. All data was generated using Igor Pro version 6.31 running on OS X/macOS.

All additional code used to further visualize and analyze the data can be found at https://gitlab.com/um-mip/mc-project-code or in the included file mc-project-code-master.zip. The Igor code was developed targeting version 7.08 running on macOS, though it may work with other versions and on other operating systems.

There also seems to be a number of tools available for both Python and R for extracting data waves from .pxp files if one does not have access to Igor (https://rdrr.io/cran/IgorR/man/read.pxp.html and https://pypi.org/project/igor/, for example). However, we have no experience with these tools and cannot speak to their usability, upkeep, etc.

Files contained:
The four zip files contain data files for each of the cells within the group. P21 = 3wk, Veh = the control mice, and all groups listed are female. mc-project-code-master.zip contains the code used to further visualize and analyze the data.
