dc.description | Generation of a conditional allele, via a sequence replacement strategy to knock-out the Aldh1a2 gene. The construct contains loxP sites that flank exon 4, a 2.5 kb 5’ short arm of homology, a 5.6 kb 3’ long arm of homology, a Diphtheria Toxin A (DTA) cassette, and a Neomycin (Neo) cassette flanked by frt sites for selective deletion. The Neo element will allow for positive selection in ES cells, while the DTA element will permit negative selection in ES cells. After homologous recombination of our conditional knock-out construct, the Aldh1a2 gene will have normal expression until Cre-mediated deletion of exon 4. Deletion of exon 4 creates a frameshift mutation and a premature stop, which should render the Aldh1a2 gene inactive. | en_US |
