Reprogramming of Steroid Metabolism in Natural Killer Cell Activation via Lipids

dc.description.abstract | Natural Killer (NK) cells are an important part of the innate immune system that primarily fight tumors and microbial infections. It has been found that obesity (i.e. an excess of lipids) impairs NK cell metabolism and cytotoxicity. In NK cells, steroids are synthesized via the mevalonate pathway, which uses acetyl-CoA as its building block and also prenylates proteins. Lipids provide a significant amount of acetyl-CoA, which may activate or deactivate the pathway. In this project, we have investigated the connection between lipid acetyl-CoA and steroid metabolism. After analyzing polymerase chain reaction (PCR) results, we found that SREBP2, an important regulator of NK cell metabolism and steroid biosynthesis, is activated by lipids. A follow-up metabolite tracing experiment showed that statins, a group of medicines that help lower cholesterol levels, lead to reprogrammed NK metabolic fluxes. This data suggests that lipid metabolism is an important determinant of NK cell function. | |
