dc.description.abstract | Dideoxynucleotide chain termination sequencing has been applied directly to genomic DNA templates by annealing radiolabeled oligodeoxynucleotide primers to unique sites in total yeast DNA and extending with avian myoblastosis virus (AMV) reverse transcriptase. The technique is used here to confirm the introduction of selectively altered tRNA genes into the Saccharomyces cerevisiae genome by gene replacement. | en_US |
